- Restriction Mapping Image Note: The ladder is lambda DNA cut with Hind III
and produces 6 visible bands (23,123 bp, 9,146 bp, 6,577 bp, 4,361 bp, 2,322 bp and 2,027 bp). The ladder is in lane 1
(closest to the ruler). The other lanes have restriction digests of lambda DNA and Eco RI performed by students.
Five bands are visible. The fifth is very light and is the farthest away from the well. Your assignment is to estimate
the fragment sizes for the restriction digest. Hint: Think about how the ruler and a graph could be used.
